Our laboratory studies the many facets of the mucosal immune response in the lungs. The focus of the laboratory is on antigen presenting dendritic cells. These cells are at the interface of innate and adaptive immunity and play a crucial role in immunoregulation and disease pathogenesis.

The first line of research deals with DC heterogeneity in the lung.  It is increasingly clear that the many functions of DCs are not shared by all subtypes of cells and an important task for the coming years is the elucidation of DC heterogeneity in the lungs.  Some cell types are important for mediating tolerance to inhaled allergens, others are important for antiviral immunity, some dictate cellular immunity, yet others control humoral immunity.  To elucidate the function of Dcs, we are employing several mouse models of disease, including house dust mite driven asthma, influenza, and embolisation of antigens into the vasculature.  This work is supported by grants from IWT, Ugent and FWO.

A second line studies the function of DCs in allergy, trying to elucidate how DCs get activated in response to allergen exposure and how they interact with bronchial epithelial cells when exposed to allergens.  For this, critical bone marrow chimeric models have been set up, as well as air liquid interface cultures of bronchial epithelial cells.  These assays are studied using cutting edge imaging techniques, such as dual photon live imaging.  In our view, understanding the precise cytokine communication between DCs, epithelial cells and T cells is vital to understanding the origin of allergy.  Increasingly, we are studyin the interaction between DCs, T cells and B cells and how this leads to regulation of IgE synthesis.  This work is supported by grants of FWO, NIH, EU (Marie Curie fellowship, IMI severe astma Network, ERS long term fellowship)

A third line studies the biochemistry and cell biology of antigen presentation.  Of the many forms of endogenous danger signals, it is increasingly clear that misfolded proteins could be important in controlling DC functions and activation.  Misfolded proteins lead to the induction of the so-called ER stress response or unfolded protein response, at the intersection of cell oxidative metabolism, glucose metabolism and activation of inflammatory pathways.  In the coming years, an important task will be to elucidate the fine regulatory mechanisms of the ER stress response in DC function and the immune response in general (including T cell and B cell function and development).  For this, important new transgenic mouse strains are being generated. A third line deals with regulation of NF-kB family members in the allergic immune response, focussing on the de-ubiquitinating enzyme A20.  Cell specific knock-outs of A20 have been made for bronchial epithelial cells, mast cells and dendritic cells in vivo, and these mice are currently being evaluated in models of asthma and influenza.  The basic aspects of antigen presentation will also be studied in these models. A fourth line deals with the immunosubversion of the immune system by tumors.  Employing a mouse model of non-small cell lung carcinoma, we are trying to identify which factors lead to a suppression of the antitumoral immune response driven by DCs.  The ultimate goal is to improve the immunotherapy of cancer using dendritic cells. Supported by IWT, Ugent  and FWO grants.

A fifth line deals with the mechanism of action of aluminium-containing adjuvants in the immune system.  These so-called alum compounds are currently one of the only clinically approaved adjuvants for use in humans.  They are also used in the mouse to stimulate development of allergy.  We are studying different chemical and structural formulations of alum, in collaboration with Oxford University. Supported by FWO.

Beschikbare onderzoekstechnieken en -infrastructuur

Our laboratory has developed several technical platforms. One of these is the set-up of a core facility for transgenesis, an initiative in which several research groups from the medical Faculty have participated (Prof. De Paepe, Prof. Leclercq, Prof. Elewaut). In 2009, the first transgnic mice were genereated and born in this unit. We offer cDNA and BAC transgene microinjections and ES-based transgenesis based on homologous recombination. A second investment has been the dual photon live imaging platform, in which fluorescently tagged cells can be followed in vivo. Finally, the flow cytometry unit now contains an LSRII 10 colour analytical flow cytometer and a high speed ARIAII cell sorter. Beginning 2010 this will be expanded by a Hercules-grant sponsored ImageStream analytical flow cytometer (Prof. Elewaut and Lambrecht).
