Eelco van Anken will present his work on endoplasmic reticulum homeostasis and how it triggers apoptosis.

Eelco van Anken (San Raffaele Scientific Institute, Milan, Italy) is our guest speaker. We developed a system that allows cells to adjust to a proteostatic insult in the endoplasmic reticulum (ER). As such we found that the unfolded protein response (UPR) senses ER stress in a ratiometric fashion based on the levels of client protein versus those of the ER chaperone BiP. Successful ER homeostatic readjustment is achieved when ultimately UPR-driven upregulation of BiP entails that its levels outmatch those of the client again. Failure in the process causes UPR sensors to negatively evaluate homeostatic readjustment and to initiate pro-apoptotic pathways based on the client/BiP ratio and on depletion levels of XBP1 mRNA.
