Septic shock is the most severe complication of sepsis, characterized by a systemic inflammatory response following bacterial infection, leading to multiple organ failure with a dramatically high mortality rate (42% at 28 days after diagnosis). Aquaporin-9 (AQP9), an aquaglyceroporin channel mainly expressed in hepatocytes and leukocytes, has been recently associated with inflammatory and infection responses. We gave evidence suggesting functional relevance of AQP9 in murine LPS-induced endotoxic shock response with possible involvement of the AQP9 channel in LPS-induced maturation of bone marrow dendritic cells (BMDC) and O2- and NO release. While work using Aqp9 knockout mice is going on to assess the precise functional implication of AQP9 in inflammatory response it is tempting to speculate that pharmacological modulation of AQP9 may reveal a novel strategy in counteracting endotoxic shock and its complications.
